Synergistic Role of Co0 and Lewis Basic Sites for Transfer Hydrogenation of Substituted Nitroarenes by In Situ Hydrogen Generation.

Designing green and efficient methods for synthesizing anilines remains a key challenge in synthetic organic chemistry. Hydrogenation of nitro compounds is one of the most significant and widely employed method for producing functionalized anilines. Consequently, designing catalytic systems capable of hydrogenating nitroarenes under environmentally benign and mild reaction conditions remains a challenge. In this regard, a highly efficient CoAl catalyst has been designed for the hydrogenation of nitroarenes to corresponding amines. Various structural, compositional, and morphological characterizations were done to understand the catalyst's structural and surface properties. The as-synthesized catalyst exhibits excellent activity for the transfer hydrogenation of 1-chloro-4-nitrobenzene to 4-chloroaniline, achieving 98% yield at 60 °C within 1.5 h in ethanol. The developed protocol demonstrated excellent activity for wide range of substituted nitroarenes. Structure-activity relationships of CoAl show that basic sites play a crucial role in the transfer hydrogenation. Furthermore, mechanistic investigations reveal that transfer hydrogenation occurs predominantly via a hydroxylamine-mediated pathway. Moreover, the catalyst maintains good catalytic activity up to four consecutive cycles. This method eliminates the need for high-pressure H2, offering a safe strategy for aromatic amine synthesis. The work demonstrates the potential of earth-abundant, non-noble-metal-based heterogeneous catalysts for hydrogenation reactions under environmentally benign conditions.

Cytokine profiles in children with PCR-confirmed HHV-6 infection according to seizure occurrence.

Although cytokines are contributors to febrile seizures, previous studies have largely involved pathogen-heterogeneous populations. Human herpesvirus 6 (HHV-6) is a common cause of febrile seizures and is useful for studying pathogen-specific inflammatory responses. This retrospective study included children with polymerase chain reaction-confirmed HHV-6 infection using stored plasma samples obtained within 72 h of hospital presentation, classified according to seizure status. Plasma cytokine levels were measured using a multiplex immunoassay. Multivariable linear regression was used to identify the cytokines associated with seizures, with Benjamini-Hochberg false discovery rate (FDR) correction across 15 cytokines. Correlations between cytokine levels and hematologic parameters were evaluated. Cerebrospinal fluid (CSF)-to-plasma cytokine ratios were analyzed. Sixty patients were included (37, without seizures; 23, seizures), with no significant differences in baseline characteristics between the groups. Plasma interleukin-8 (IL-8) showed an association with seizures (adjusted fold change 1.84, p = 0.03), but this association did not remain statistically significant after FDR correction (q = 0.45). Inverse correlations between IL-8 (and to a lesser extent macrophage inflammatory protein-1 alpha) and neutrophil and platelet counts were observed in the non-seizure group only. In the paired analyses (n = 8), the CSF-to-plasma ratios of IL-8 and monocyte chemoattractant protein-1 exceeded 1.0, with a trend toward higher IL-8 ratios in the seizure group after adjusting for age. Although exploratory, findings from this pathogen-homogeneous cohort suggest that IL-8 is a candidate cytokine associated with both seizure occurrence and an altered relationship with neutrophil and platelet counts, warranting further study.

AMBMP Hydrochloride Reverses STZ-Induced Alzheimer-Type Dementia Associated with Wnt/β-catenin/TLR4 Signaling.

Alzheimer's disease (AD) is a debilitating neurodegenerative disorder with progressive cognitive decline and neuronal loss. This study investigated the neuroprotective effects of AMBMP Hydrochloride, a Wnt/β-catenin agonist, in a streptozotocin (STZ)-induced mice model of AD, elucidating the interplay between dysregulated Wnt/β-catenin signaling and Toll-Like Receptor 4 (TLR4)-mediated inflammation, with Palmitic acid used as a TLR4 pathway modulator. Mice received bilateral intracerebroventricular (i.c.v.) injections of STZ (3 mg/kg) on Day 1 and Day 3, followed by administration of Donepezil (3 mg/kg)/ AMBMP Hydrochloride (5 mg/kg and 10 mg/kg)/Palmitic acid (TLR4 agonist, 20 mg/kg) via the intraperitoneal (i.p.) route from Day 4 to Day 22. STZ-treated mice exhibited significant cognitive dysfunction, characterized by impaired performance in the Morris Water Maze (MWM) task along with increase in acetylcholinesterase (AChE) activity, oxidative stress (thiobarbituric acid reactive substances; TBARS), neuroinflammation [tumor necrosis factor alpha (TNF-α)/Interleukin-6 (IL-6)/Interleukin-1 beta (IL-1β) and nuclear factor kappa B (NF-κB)] and decreased reduced glutathione (GSH) levels. However, AMBMP Hydrochloride significantly improved behavioral and biochemical alterations possibly through modulation of Wnt/β-catenin signaling and attenuation of TLR4-mediated inflammation. Interestingly, Palmitic acid co-treatment was found to counteract these protective effects, further pointing to a role of TLR4 in the pharmacological effect of AMBMP Hydrochloride. In summary, we confirmed that AMBMP Hydrochloride exhibits potent neuroprotective effects associated with modulating Wnt/β-catenin/TLR4 signaling, offering a promising therapeutic approach against Alzheimer-Type Dementia. Future studies should delve deeper into the molecular mechanisms and translational promise of AMBMP hydrochloride, paving the way for its development as a potential candidate for AD management.

Methomyl resistance in Musca domestica: mechanisms and inheritance.

Methomyl resistance is a documented problem in Musca domestica field strains worldwide, including Pakistan, where the insecticide is commonly used. To address this, a methomyl-resistant near-isogenic line (Meth-R) was created specifically to determine its resistance inheritance mode and cross-resistance profile. Analysis of reciprocal F1 and F1' crosses demonstrated autosomal inheritance of methomyl resistance in the Meth-R strain, as evidenced by similar median lethal dose (LD50) values and the absence of sex linkage or maternal effects. Resistance expression was incompletely dominant, with dominance values of 0.7 (F1) and 0.6 (F1'). Significant deviations between observed and expected mortality in self-bred (F2 and F2') and backcross (BC1-BC4) progenies (chi-square analyses) indicate resistance is likely controlled by multiple genes. The Meth-R strain showed no cross-resistance to alpha-cypermethrin, fluralaner, pirimiphos-methyl, or clothianidin. Additionally, observing significant synergism of methomyl with both piperonyl butoxide (PBO; oxidase inhibitor) and S,S,S-tributyl phosphorotrithioate (DEF; esterase inhibitor) implicates metabolic detoxification as a key mechanism of methomyl resistance. The Meth-R strain exhibits methomyl resistance inherited as an autosomal, incompletely dominant trait under polygenic control. Although there was cross-resistance in the Meth-SEL strain to alpha-cypermethrin, fluralaner, pirimiphos-methyl, and clothianidin versus the lab susceptible strain, selection with methomyl did not increase the level of cross-resistance. These results offer a foundation for combating methomyl resistance.

Translation and validation of the extended version of the effort assessment scale for adults with cochlear implant in Turkish language.

Although assessing listening effort is clinically essential, no validated Turkish self-report measure currently exists. This study aimed to translate and culturally adapt the Extended version of the Effort Assessment Scale (EEAS) into Turkish and to evaluate the validity and reliability of the Turkish EEAS (T-EEAS) in adults with typical hearing (TH) and cochlear implant (CI) users. A standardized cross-cultural adaptation protocol was followed. Psychometric properties were evaluated in 567 adults (463 TH and 104 CI users). Construct validity was examined using confirmatory factor analysis. Reliability was assessed using Cronbach's alpha, test-retest correlation, intraclass correlation coefficient (ICC), and Bland-Altman agreement analysis. Discriminant validity was evaluated by comparing T-EEAS scores between groups. Confirmatory factor analysis supported a two-factor structure with good model fit. The T-EEAS showed high internal consistency (Cronbach's alpha = 0.938) and excellent test-retest reliability (Spearman's r = 0.867; ICC = 0.877). CI users reported higher listening effort than the TH group (P < 0.001), with a large effect size (Cliff's δ = 0.838). The T-EEAS captures subjective listening effort in Turkish-speaking populations. The two-factor structure distinguishes effort in quiet versus noise, which may support outcome monitoring in audiological rehabilitation. The higher scores observed in CI users support the discriminant validity of the scale. The T-EEAS is a valid and reliable measure of subjective listening effort in Turkish-speaking adults and can distinguish between TH adults and CI users, supporting its use in clinical and research settings.

Microbial Communities Vary by Body Region but Remain Consistent Across Color Morphs in the Solitary Ascidian Rhopalaea abdominalis.

Ascidians are filter-feeding marine invertebrates that display inter- and intraspecific color variation and host diverse microbiomes. Although color variation has been used as an indicator of speciation and correlated with distinct microbiome structure in some colonial species, there is limited research on microbiome variation across color morphs and body regions in solitary ascidians. To address these knowledge gaps, three color morphs (purple, orange, and intermediate shades of pink) of the solitary Belizean ascidian Rhopalaea abdominalis were collected for phylogenetic analysis using the Cytochrome Oxidase I (COI) gene and for microbial characterization via sequencing the V4 region of the 16S rRNA gene. Microbial sequences derived from the ascidian branchial sac, gut, and tunic body regions were processed using both operational taxonomic unit (OTU) and amplicon sequence variant (ASV) pipelines to determine the resolution and consistency of sequence processing methods using mothur and QIIME 2 software, respectively. COI sequencing revealed two clades that were independent of color morphs. Microbiome characterization showed no significant differences in alpha- or beta-diversity across ascidian color morphs or COI clades but distinct microbiomes within each body region. The branchial sac harbored a higher number of core members (i.e., detected across all host individuals) that included known ascidian symbionts (e.g., Endozoicomonas), whereas the gut and tunic were colonized by taxa identified in seawater and across multiple classes of marine invertebrates. Results were highly congruent between OTU- and ASV-based pipelines, yielding statistically equivalent alpha-diversity metrics, consistent beta-diversity patterns, and similar core membership profiles. These findings reinforce the validity of both methods for studying microbial symbiont communities and corroborate previous research showing distinct microbiomes by body region and overall conservation when color morphs lack genetic differentiation.
