The link between low maternal cholesterol levels and increased risk of fetal patent ductus arteriosus and septal defects: Large-scale cohort retrospective study.

To investigate whether low maternal cholesterol levels during pregnancy are associated with an increased risk of neonatal Patent Ductus Arteriosus (PDA) and Atrial or Ventricular Septal Defect (ASD/VSD). Medical records of 36,633 pregnant women who received care and delivered at Fujian Maternity and Child Health Hospital, China, from 2016 to 2020 were retrospectively analyzed. Maternal serum lipid levels were measured one week before delivery, and Congenital heart disease (CHD) was diagnosed by echocardiography within three days of birth. Logistic regression models were adjusted for neonatal sex, maternal age, gestational diabetes and hypertension, gravidity, parity, and preterm birth. Among 36,633 mother-child pairs, 906 (2.5%) cases of CHD were identified. Lower maternal TC, LDL-C, and HDL-C levels were significantly associated with increased CHD risk. Each 1 mmol/L increase in TC was associated with an 11% decrease in CHD risk (OR: 0.89, 95% CI 0.85, 0.95). LDL-C showed a similar protective effect (OR: 0.88, 95% CI 0.82, 0.95), while HDL-C demonstrated the strongest association (OR: 0.61, 95% CI 0.51, 0.73). The relationship between TG and CHD risk was not significant after adjustment (OR: 0.97, 95% CI 0.92, 1.02; p=0.224). For PDA specifically, TC, LDL-C, and HDL-C remained significantly protective; for ASD/VSD, only TC demonstrated significance (OR: 0.91, 95% CI 0.83, 0.99). TG showed no significant association with any CHD subtype. This study indicates that lower maternal levels of TC, LDL-C, and HDL-C may be significantly associated with an increased risk of PDA, while lower TC levels could be associated with ASD/VSD risk. The relationship between maternal TG levels and CHD could not be demonstrated. Future prospective multicenter and intervention studies are needed to validate the results.

Race Performance Following Arthroscopic Surgery for Femoropatellar and Tarsocrural Osteochondrosis in Juvenile Thoroughbreds.

The objective of this study is to evaluate racing performance of Thoroughbred yearlings following arthroscopic surgery for osteochondrosis (OC) of the femoropatellar (FP) and tarsocrural (TC) joints. This is a retrospective study of Thoroughbreds < 2 years old undergoing arthroscopic surgery for OC in the FP or TC joint (2010-2022). For each case, two age- and sex-matched controls from the same stud farm were selected. Lesions were graded on preoperative radiographs and described. Racing outcomes (number of starts, wins, places, podiums, prize money at 2nd, 3rd, and ≥4-year-old seasons, and career duration) were compared between groups using parametric or nonparametric tests according to data distribution (Student's t-test/analysis of variance or Mann-Whitney U/Kruskal-Wallis tests). A total of 49 horses included: 27 in the FP group and 22 in the TC group. In the FP group, the lateral trochlear ridge was most often affected; 14 were bilateral, two had OC lesions in another joint. In the TC group, the distal intermediate ridge of the tibia was mainly affected; five were bilateral and one had OC lesion in another joint. No significant differences were found between cases and controls in number of starts, wins, places, podiums, and prize money earned at 2nd, 3rd, and ≥4-year-old racing careers or career duration for either group (p > 0.05). In both groups, no significant performance differences were found between unilateral and bilateral cases (p > 0.05). Racing performance after FP or TC OC surgery was comparable to that of unaffected controls. Bilateral cases performed as successfully as unilateral.

Global DNA methylation and hypertension control Vis-À-Vis lipids: an exploratory population based study from rural Punjab, India.

Hypertension remains a major global health challenge, accounting for a substantial burden of mortality and morbidity. In India, despite advances in pharmacological management, optimal blood pressure (BP) control is still not achieved in a significant proportion of the population. Abnormal lipid profiles frequently coexist with hypertension, synergistically increasing the risk of cardiovascular diseases. Emerging evidence suggests that DNA methylation may mediate the effects of metabolic stressors on gene regulation, contributing to the pathophysiology of hypertension and related cardiometabolic disorders. This population-based study investigates the interrelationship between blood pressure, lipid profiles, and global 5-methylcytosine (5mC) levels across normotensive individuals, hypertensives identified by the investigators and not yet on medication, and hypertensives on medication (both controlled and uncontrolled). A total of 900 individuals (450 hypertensive cases and 450 age- and sex-matched controls) were recruited from rural Punjab. Anthropometric parameters (BMI, WC, HC), BP, lipid levels, and global DNA methylation were assessed using standardized protocols. Data were analyzed using SPSS v22. Findings revealed that abnormal levels of triglycerides (TG), total cholesterol (TC), and low-density lipoprotein cholesterol (LDL-C) were significantly higher among hypertensive individuals on medication with uncontrolled BP. These individuals also exhibited significantly lower global 5mC levels, indicating hypomethylation. The results suggest that a high lipid burden may not only compromise the efficacy of antihypertensive medication but also contribute to global DNA hypomethylation, which is often associated with accelerated biological aging and elevated cardiovascular risk. The study highlights the importance of lipids not in hypertension control but also in maintenance of Global DNA methylation.

Anti-inflammatory, wound healing, and hepatoprotective effects of Acampe papillosa (Lindl.) Lindl. seed in wistar rats: insights from biochemical, histopathological, and in silico investigations.

Acampe papillosa (Lindl.) Lindl. has long been used to cure a variety of illnesses, including rheumatism and traumatic tissue injuries. Nevertheless, there is a dearth of comprehensive information on its phytochemical composition and pharmacological actions. Therefore, here, we present the first thorough pharmacological and histopathological analysis of Acampe papillosa seed methanolic extract (ME-APS), together with in silico molecular docking studies, hepatoprotective, wound-healing, and anti-inflammatory properties. Before pharmacological evaluations, safety was assessed through oral and dermal toxicity studies. The anti-inflammatory activity of ME-APS was tested using carrageenan and formalin-induced paw oedema models, and the wound healing potential was observed using incision and excision wound models. Hepatoprotective activity was studied in the CCl4-induced hepatotoxicity model in rats. In oral (up to 5000 mg/kg) and dermal (10% ointment formulation) toxicity studies, ME-APS was found to be safe with no visible signs of toxicity. In the study on anti-inflammatory properties, ME-APS significantly reduced paw oedema in carrageenan and formalin models of inflammation. Wound healing assays showed that the extract significantly improved wound contraction, tensile strength, epithelialization, collagen deposition, and fibroblast proliferation in a dose-dependent way. The hepatoprotective study showed that ME-APS treatment significantly restored the altered biochemical parameters, such as ALT, AST, TC, TG, and LDL levels, and ameliorated CCl4-induced histopathological liver damage. Further, the molecular docking study showed that the major phytochemicals in the extract, such as squalene, cholesta-4,6-dien-3-ol (3β), stigmasterol, and γ-sitosterol, showed significant binding affinities for COX-2 and TGF-β-related targets involved in inflammation, wound healing, and hepatoprotection. Additionally, a high probability of biological efficacy was predicted using PASS (Prediction of Activity Spectra for Substances) analysis. Overall, the findings suggest that ME-APS has possible anti-inflammatory, wound healing, and hepatoprotective activities that support its traditional medicine use and imply that it is a potential source of therapeutic phytochemicals.

Predictive modelling of tetracycline removal by I-Bi/Bi2WO6/MWCNTs photocatalyst using RSM and ANN-PSO hybrid machine learning approach.

A visible-light responsive heterostructure photocatalyst (I-Bi/Bi₂WO₆/MWCNTs) was successfully synthesized using an ethylene glycol-assisted hydrothermal method for the photodegradation of the tetracycline (TC) antibiotic. To analyze and streamline the process performance, a hybrid machine learning model that incorporated artificial neural networks (ANN) with particle swarm optimization (PSO) and response surface methodology (RSM) was applied to forecast and optimize key operating parameters, such as solution pH, initial TC concentration, contact time, and photocatalyst dosage. The 4-8-1 topology of ANN-PSO was found to be the optimal network, and the prediction model of TC removal was demonstrated as a matrix of explicit equations. The R² of randomized training (0.98), testing (0.99), and validation (0.96) at the optimized topology confirms the efficiency of the developed ANN-PSO model. The ANN-PSO model showed a better predictive performance in comparison with the RSM model (R² = 0.960, RMSE = 5.379), with the correlation coefficient (0.981) being higher and the RMSE (4.712) lower. A 96.8% TC removal was achieved at solution pH 8, initial TC concentration of 20 mg/L, contact time of 160 min, and photocatalyst dosage of 25 mg.
