Synergistic Role of Co0 and Lewis Basic Sites for Transfer Hydrogenation of Substituted Nitroarenes by In Situ Hydrogen Generation.

Designing green and efficient methods for synthesizing anilines remains a key challenge in synthetic organic chemistry. Hydrogenation of nitro compounds is one of the most significant and widely employed method for producing functionalized anilines. Consequently, designing catalytic systems capable of hydrogenating nitroarenes under environmentally benign and mild reaction conditions remains a challenge. In this regard, a highly efficient CoAl catalyst has been designed for the hydrogenation of nitroarenes to corresponding amines. Various structural, compositional, and morphological characterizations were done to understand the catalyst's structural and surface properties. The as-synthesized catalyst exhibits excellent activity for the transfer hydrogenation of 1-chloro-4-nitrobenzene to 4-chloroaniline, achieving 98% yield at 60 °C within 1.5 h in ethanol. The developed protocol demonstrated excellent activity for wide range of substituted nitroarenes. Structure-activity relationships of CoAl show that basic sites play a crucial role in the transfer hydrogenation. Furthermore, mechanistic investigations reveal that transfer hydrogenation occurs predominantly via a hydroxylamine-mediated pathway. Moreover, the catalyst maintains good catalytic activity up to four consecutive cycles. This method eliminates the need for high-pressure H2, offering a safe strategy for aromatic amine synthesis. The work demonstrates the potential of earth-abundant, non-noble-metal-based heterogeneous catalysts for hydrogenation reactions under environmentally benign conditions.

[Utility of global coagulation assays for hemostasis management in hyperclearance-type autoimmune acquired coagulation factor V deficiency].

We report the case of an 80-year-old woman in which marked prolongation of prothrombin time (PT) and activated partial thromboplastin time (APTT) noted upon diagnosis of autoimmune hemolytic anemia revealed severe factor V (FV) deficiency. A mixing study showed a downward-convex pattern and the Bethesda assay for inhibitors was negative, which initially suggested congenital FV deficiency. Although the patient had concomitant early-stage esophageal cancer, infusions of fresh frozen plasma (FFP) failed to correct the coagulopathy, necessitating postponement of endoscopic therapy. Subsequently, a comprehensive evaluation by the Standardization Committee of the Japanese Ministry of Health, Labour and Welfare detected anti- FV IgG, establishing the diagnosis of hyperclearance-type autoimmune FV deficiency (AiFVD). Despite corticosteroid therapy, conventional coagulation screening test results remained markedly abnormal, complicating periprocedural management. In contrast, viscoelastic testing (VET) and thrombin generation testing (TGT), including ex vivo supplementation assays, indicated that adequate hemostasis could be achieved, allowing the endoscopic treatment to proceed safely. This case underscores that, even in FV deficiency with a negative inhibitor screen, testing for anti-FV IgG is essential for identifying hyperclearance-type AiFVD. Furthermore, ex vivo VET and TGT provided actionable assessments of hemostatic capacity and proved useful for guiding management in AiFVD.

Telomeres are repetitive DNA sequences at the ends of linear chromosomes bound by specialized proteins. In our previous quantitative proteomics screen for telomere-binding proteins of Caenorhabditis elegans, we identified DVE-1, a homolog of mammalian SATB proteins and a transcription factor, as a telomere repeat-binding protein. Here, we validate DVE-1 as a telomere-binding protein in C. elegans, demonstrating in vitro binding of DVE-1 to the single-stranded C-rich telomeric sequence and in vivo co-localization with the telomere-binding protein POT-1. RNA interference-mediated knockdown of dve-1 resulted in reduced TERRA expression and enhanced compaction of telomeric chromatin. Subsequent transcriptomic and proteomic analyses suggest a role for DVE-1 in the regulation of telomeric chromatin organization. Finally, DVE-1 immunoprecipitation followed by mass spectrometry revealed all the core components of the nucleosome remodeling and deacetylase (NuRD) complex as interaction partners, implicating DVE-1 in the coordination of NuRD complex activity in the context of telomere organization.

Emergence of a novel NDM-4-producing Citrobacter freundii complex species in a complicated urinary tract infection treated with cefiderocol.

Multidrug-resistant (MDR) Gram-negative infections represent a growing global health threat with increasingly limited therapeutic options. We report a case of complicated urinary tract infection in a 70-year-old woman with underlying renal cysts complicated by rupture and subsequent formation of left infrarenal and paracolic pelvic abscesses. The initial infection was caused by Escherichia coli; however, following initial prolonged broad-spectrum antimicrobial therapy, a novel New Delhi metallo-β-lactamase (NDM type carbapenemase)-producing Citrobacter species was subsequently isolated from urine cultures. Whole-genome sequencing confirmed identification as a novel Citrobacter species and identified the presence of a blaNDM-4 gene. Antimicrobial susceptibility testing demonstrated resistance to most tested agents, with susceptibility limited to nitrofurantoin, cefiderocol, aztreonam, doxycycline, and tigecycline. Repeat abscess drainage and treatment with cefiderocol resulted in clinical and microbiological resolution of infection. This case underscores the critical role of drainage and access to newer antibiotic agents in management of complex intra-abdominal infections. Genome sequencing aids the detection of novel reservoirs of antibiotic resistance genes in carbapenemase-producing Enterobacterales that may emerge during prolonged antimicrobial exposure.

Psychometric properties and age- and gender-specific normative values of the Patient Health Questionnaire-4 among older adults aged 60 years and above.

Mental health problems in older adults are often underrecognized despite their substantial impact on health and well-being. Brief, valid screening tools with age-specific normative data are needed for this growing population. This study examined the psychometric properties of the Patient Health Questionnaire-4 (PHQ-4) and established age- and gender-specific normative values for adults aged 60 years and older in Germany. Data were drawn from four nationally representative face-to-face household surveys conducted in Germany between 2021 and 2024. Four of the total 3,212 participants were excluded due to missing PHQ-4 responses, resulting in a final sample of N = 3,208. The psychometric properties of the Generalized Anxiety Disorder-2 (GAD-2), the Patient Health Questionnaire-2 (PHQ-2), and the Patient Health Questionnaire-4 (PHQ-4) were evaluated. The PHQ-4 showed excellent internal consistency. The correlated two-factor model, distinguishing depression and anxiety symptoms, demonstrated excellent fit. Measurement invariance was supported across gender and age groups. Women reported higher overall psychological distress than men, mainly due to higher anxiety symptoms, and PHQ-4 scores were highest in the oldest age group. Convergent validity was supported by moderate to strong negative correlations with quality of life, resilience, sense of coherence, and well-being. Age- and gender-specific normative values were provided for the PHQ-2, GAD-2, and PHQ-4. The PHQ-4 is a reliable and valid ultra-brief screening instrument for psychological distress. The age- and gender-specific normative values provided here may improve score interpretation for community-dwelling older adults in Germany and help identify individuals who may require further clinical assessment.

AMBMP Hydrochloride Reverses STZ-Induced Alzheimer-Type Dementia Associated with Wnt/β-catenin/TLR4 Signaling.

Alzheimer's disease (AD) is a debilitating neurodegenerative disorder with progressive cognitive decline and neuronal loss. This study investigated the neuroprotective effects of AMBMP Hydrochloride, a Wnt/β-catenin agonist, in a streptozotocin (STZ)-induced mice model of AD, elucidating the interplay between dysregulated Wnt/β-catenin signaling and Toll-Like Receptor 4 (TLR4)-mediated inflammation, with Palmitic acid used as a TLR4 pathway modulator. Mice received bilateral intracerebroventricular (i.c.v.) injections of STZ (3 mg/kg) on Day 1 and Day 3, followed by administration of Donepezil (3 mg/kg)/ AMBMP Hydrochloride (5 mg/kg and 10 mg/kg)/Palmitic acid (TLR4 agonist, 20 mg/kg) via the intraperitoneal (i.p.) route from Day 4 to Day 22. STZ-treated mice exhibited significant cognitive dysfunction, characterized by impaired performance in the Morris Water Maze (MWM) task along with increase in acetylcholinesterase (AChE) activity, oxidative stress (thiobarbituric acid reactive substances; TBARS), neuroinflammation [tumor necrosis factor alpha (TNF-α)/Interleukin-6 (IL-6)/Interleukin-1 beta (IL-1β) and nuclear factor kappa B (NF-κB)] and decreased reduced glutathione (GSH) levels. However, AMBMP Hydrochloride significantly improved behavioral and biochemical alterations possibly through modulation of Wnt/β-catenin signaling and attenuation of TLR4-mediated inflammation. Interestingly, Palmitic acid co-treatment was found to counteract these protective effects, further pointing to a role of TLR4 in the pharmacological effect of AMBMP Hydrochloride. In summary, we confirmed that AMBMP Hydrochloride exhibits potent neuroprotective effects associated with modulating Wnt/β-catenin/TLR4 signaling, offering a promising therapeutic approach against Alzheimer-Type Dementia. Future studies should delve deeper into the molecular mechanisms and translational promise of AMBMP hydrochloride, paving the way for its development as a potential candidate for AD management.
