Effect of Alkalization With Potassium Carbonate and Ammonium Bicarbonate on the Chemical and Physical Properties of Cocoa Butter: A Factorial and Central Composite Design Approach.

Sadeghzadeh Afghan A, Taghinezhad Maryam M, Pirsa Sajad S

This study investigated the effects of cocoa butter alkalization using potassium carbonate and ammonium bicarbonate (0%-6%) on acidity, peroxide value, iodine value, saponification value, moisture content, slip melting point, pH, and lead content. A factorial design and central composite design (CCD) were employed. The highest peroxide value (0.1 meq O2/kg) was observed in samples containing 3% potassium carbonate +3% ammonium bicarbonate and 6% potassium carbonate +6% ammonium bicarbonate. The highest acidity was related to the control sample (1.77), while the lowest was found in the sample with 6% of both alkalis (0.77). The control sample also showed the lowest saponification value (189.61 mg KOH/g fat), whereas the highest was recorded for the 6% + 6% sample (199.04 mg KOH/g fat). The highest iodine value (35.91 g I2/100 g fat) belonged to the 6% + 6% sample, and the lowest (32.42 g I2/100 g fat) to the sample containing 6% ammonium bicarbonate alone. Minimum moisture content (0.053%) was observed in the sample with 6% potassium carbonate +3% ammonium bicarbonate, and maximum (0.077%) in the sample with 3% + 3%. The lowest pH (5.05) was for the control, and the highest (9.87) for the 6% + 6% sample. The highest slip melting point (30.23°C) was recorded for the control, and the lowest (26°C) for the sample with 3% potassium carbonate +6% ammonium bicarbonate. All samples complied with Iranian national standards for lead content. The results indicated that 3% potassium carbonate alone, 3% ammonium bicarbonate alone, and 3% potassium carbonate +6% ammonium bicarbonate are suitable for cocoa alkalization. Finally, mathematical models were developed using response surface methodology to establish quantitative relationships between the chemical properties of cocoa butter and the concentrations of the alkaline agents, enabling the achievement of desired product characteristics.

Ancient Endorheic Lake Basins: Promising Sampling Sites for the Tianwen-3 Mars Sample Return Mission.

Li Yi-Liang YL, Wu Bo B

The China National Space Administration has announced its Mars Sample Return mission Tianwen-3 to search for life on Mars. Tianwen-3 will be launched in 2028 and return ∼500 g of samples in 2031. Since the Tianwen-3's stationary lander will use a built-in 2 m drill to retrieve samples from depth and a robotic arm to scoop surface deposits, the strategy for selecting the landing site should differ from that of missions with rovers. Based on discoveries by the Curiosity and Perseverance rovers, we suggest that Tianwen-3 should land in an ancient endorheic lake basin where aqueous sediments and rich authigenic minerals are preserved due to the past presence of a stable body of water and high likelihood of a sustained period of vertical cycling of deep fluids. Our recent observations of brines and iron oxides precipitated by iron bacterial metabolism in a hyperarid, highly evaporative Mars analog basin further support the hypothesis that low-lying, closed crater basins on Mars are of high astrobiological value for upcoming sample return missions.

Uniaxial Strain-Driven Ferroelastic Domain Tuning in LaAlO 3.

Roeper Matthias M, Buschbeck Robin R, Wetzel Jakob J, Ritschel Tobias T et al.

Multiferroic domain walls in functional oxides exhibit properties distinct from the bulk and are increasingly exploited as active elements in nanoelectronic and photonic devices. Deterministic tuning of domain populations has typically remained limited to local solutions, or removal with temperature. Here, the manipulation of ferroelastic domain structure, and removal of the twin walls between them in single-crystal LaAlO 3 using in situ uniaxial strain is demonstrated. Combining atomic force microscopy, X-ray diffraction, and Raman spectroscopy with first-principles calculations, the microscopic evolution of the twin domain population is mapped through a strain-driven transition from the rhombohedral R 3 ¯ c ground state toward the predicted orthorhombic F m m m phase. Applied strains below 0.5 % produce pronounced surface flattening and large-scale domain reorganization, establishing uniaxial strain as a technically accessible tuning parameter for ferroelastic domain engineering. These results open a route to active alterations of domain architectures in LaAlO 3 -based heterostructures, with implications for epitaxial film properties, nanoscale phonon-polariton optics, and ultrafast lattice control.

Proximity of pre-mRNA 3' end processing and transcription termination is associated with enhanced gene expression.

Stępień Agata A, Pulimamidi Deepshika D, Plens-Gałąska Martyna M, Lalonde Maxime M et al.

Two key factors required for pre-mRNA 3' end processing, CPSF73 and PCF11, are linked to poor cancer prognosis, yet their oncogenic roles remain unclear due to their dual function in transcription termination. Using a colorectal cancer (CRC) progression model, we show that primary tumor cells depend on high CPSF73 and PCF11 levels, whereas metastatic cells are less sensitive. This dependency is not explained by alternative polyadenylation (APA) or transcription-replication conflicts. Instead, we identify an uncoupling of 3' cleavage and transcription termination during CRC progression. Primary tumor cells exhibit a global shift toward proximal termination alongside distal APA usage, while metastatic cells partially revert to normal termination patterns and favor proximal APA. Notably, the distance between 3' cleavage and termination is nearly halved in primary tumor cells compared with normal cells and becomes particularly short in oncogenes. Reduced cleavage-termination distance correlates with increased gene expression both within and across cell states. These findings identify a previously unrecognized link between transcription termination architecture and gene expression regulation.

Global conservation area coverage substantially exceeds current estimates.

O'Neill Helen M K HMK, Beeken Joe J, Bingham Heather C HC, Golden Kroner Rachel E RE et al.

Conservation areas are critical for tackling the global environmental crisis1. These sites are reported as covering 18.5% of the terrestrial realm2 and governments have committed through Global Biodiversity Framework Target 3 to increase this to at least 30% by 2030 (ref. 3). There are concerns that this potential near-doubling of conservation area coverage will negatively affect vulnerable people and over-stretch conservation resources4. However, nationally available datasets indicate that more conservation areas exist than are reflected in global figures5-7. We evaluated progress towards the area component of Target 3 using a globally representative sample across 25 countries and territories that incorporated these national datasets8. Extrapolating from this sample, we show that conservation areas likely already cover 26.81% of the terrestrial realm. We may be much closer to achieving 30% terrestrial coverage than previously thought. However, we also show that the extra land does not substantially address gaps in ecological representativeness, indicating that solely meeting the area component of Target 3 will do little to solve continuing global biodiversity loss. More focus and resources are needed to achieve the other components of the target, using a more strategic and inclusive approach to fill present gaps and produce equitable and effective conservation area networks.

Automated Segmentation of Retinal Pigment Epithelium Layers in High-Resolution OCT.

Ritter Giulia G, Isselmann Ben B, Saßmannshausen Marlene M, Jauch Anna S AS et al.

To quantify retinal pigment epithelium (RPE) layer thicknesses and reflectivity in healthy participants across different age groups using improved axial, high-resolution (High-Res) OCT. A cross-sectional observational study. A total of 110 eyes from 110 healthy participants were included. Participants were categorized into 3 age groups: group 1 (<40 years), group 2 (40-65 years), and group 3 (>65 years). High-resolution OCT imaging was performed, followed by the segmentation of 3 RPE layers. The inner layer corresponded to RPE layer 1 (High-Res OCT nomenclature) and outer segment interdigitation zone 2, the middle RPE layer corresponded to RPE layers 2 and 3, and the outer RPE layer corresponded to RPE layers 4 and 5 and Bruch membrane. A convolutional neural network (CNN) was trained for automated RPE layer recognition. Volumetric thickness maps and reflectivity values were generated to assess age-related differences across the macula and analyzed using the Kruskal-Wallis and Mann-Whitney U tests. The primary outcome measures were RPE thicknesses and vitreous-normalized reflectivity of the 3 RPE layers across the 3 age groups. The CNN achieved high segmentation accuracy with a mean Dice of 0.85. Hyporeflective RPE layer 1 was absent at the fovea but increased in thickness with eccentricity. Significant age-related increases were observed in the reflectivity throughout the middle hyperreflective RPE layer, at the fovea (H = 13.8; P = 0.001), the inner ETDRS ring (H = 19.8; P < 0.001), and the outer ETDRS ring (H = 16.2; P < 0.001). For the outer hyperreflective RPE layer, an increase in thickness with age was noted in the outer (H = 8.63; P = 0.013), along with an increased reflectivity at the fovea (H = 14.8; P < 0.001) and the inner ETDRS ring (H = 14.35; P < 0.001). OCT imaging with improved resolution enables detailed segmentation of RPE layers, revealing moderate but significant age-related changes in reflectivity and thickness. These findings provide normative data essential for identifying early retinal alterations and developing structural biomarkers for age-related retinal diseases.
