October 1, 2026: Evommune, Inc. (NYSE: EVMN) reported additional Phase 2a proof-of-concept data for EVO301, an IL-18 inhibitor, in 70 adults with moderate-to-severe atopic dermatitis at the EADV 2026 Congress. In the trial, patients received IV EVO301 5 mg/kg at baseline and week 4 (n=48) or placebo (n=22), with the primary endpoint at week 12.
EVO301 met the primary endpoint, with a 55% least-squares mean reduction in EASI at week 12 versus 22% for placebo (p<0.01). Separation from placebo was seen at week 4 (41% vs. 18%) and week 8 (50% vs. 16%). At week 12, EASI-50 was 63% for EVO301 versus 23% for placebo, and EASI-75 was 29% versus 9%.
Biomarker analyses showed reductions in Th2 markers CCL-17/TARC and CCL-22, including about 75% reductions in patients with high baseline TARC, plus reductions in Th1, Th17 and Th22 markers including IL-22. Safety was favorable: no treatment-related serious or severe adverse events, no discontinuations due to adverse events, no clinically significant lab abnormalities, and no conjunctivitis; treatment-related adverse events were 10.4% with EVO301 versus 13.6% with placebo.
The company said it plans to start a global subcutaneous Phase 2b dose-ranging trial in mid-2027.
