May 6, 2026: Xencor, Inc. (NASDAQ: XNCR) reported Q1 2026 results and provided pipeline catalyst timelines across oncology and autoimmune programs.

XmAb819 (ENPP3 x CD3) in advanced clear cell renal cell carcinoma (ccRCC): expansion cohort data from the ongoing Phase 1 study will be presented at a medical conference in 2H 2026 to help define a recommended Phase 3 target dose; a pivotal study is planned for 2027. A ccRCC sub-study in patients not previously treated with a tyrosine kinase inhibitor is planned to start in 3Q 2026. Expansion cohorts in ENPP3+ colorectal cancer, NSCLC, and papillary RCC are open.

XmAb541 (CLDN6 x CD3) in advanced gynecologic and germ cell tumors: Phase 1 dose-escalation data presentation planned for 2H 2026; pivotal-study planning to be evaluated during 2027. Xencor is initiating a combination study of XmAb541 with XmAb808 (B7H3 x CD28) in CLDN6+ high-grade serous ovarian cancer.

XmAb942 (anti-TL1A) in ulcerative colitis: global Phase 2b XENITH-UC enrolling; blinded interim analysis expected around year-end 2026 and primary endpoint (week-12 clinical remission by modified Mayo score) analysis expected in 2H 2027. Final Phase 1 healthy-volunteer results were presented at DDW May 2026 (well tolerated; estimated 74-day half-life; no ADA impact reported).

XmAb412 (TL1A x IL23p19): first-in-human healthy participant study planned to start in 3Q 2026; preclinical data presented at DDW May 2026.

Plamotamab (CD20 x CD3) in RA and XmAb657 (CD19 x CD3) in idiopathic inflammatory myopathies: Phase 1/1b progress updates planned for 2H 2026. Management will present at the BofA Securities Health Care Conference on May 12, 2026.
